I had actually done a similar deal tonight using two of the Culturelle and they refused to give me the gift card because they said the Cartwheel made it invalid. Even the manager said they wouldn’t do a gift card using Cartwheels on this deal (but they gave me the $10 gift card for buying the three Charmin toilet papers and using the Cartwheel on those!) because it took the total below $25.

I don’t know if it was just a one time thing at my store (never had issues using cartwheels on minimum amount purchases before) or if they’re trying out a new policy, but YMMV on this.

My target allowed the gift card and cartwheel. I had no problem at all with this deal. The cashier did not even question the transaction. My target is located in Arizona.
