i went in-store to buy Up &Up diapers this earlier this week, and it was not marked on shelf, not listed in ad, and at the register the gift card did not get triggered.

Those are both threshold coupons (I.e. spend$X and get $Y back) so, per target coupon policy, in order to apply both coupons in one transaction you would have to meet the threshold for each offer separately (in your case that means spending $150). H2H.
